By - is used state who or what is doing the action; or how an action is done

  • I go to school by car.
  • We are going to visit my sister by bus.
  • I want to travel though Europe by bike.
  • The girls are going to the store by foot.

On - is used when something is touching something; usually when something or someone is touching the top of something or someone

  • My mother puts me on the bus to go to school.
  • I do not like to sit on an airplane for a long time.
  • How many people are on the bus?
